HGF reduces inflammatory cytokine and cytolytic enzyme production by activated CD8 + T cells. Pmel-1 splenocytes were cultured for 5 days in IL-2 alone or in combination with HGF, following antigen-specific stimulation. (a and b) Intracellular cytokine staining of day 5-cultured CD8+ T cells showed that addition of HGF not only decreased (a) the number of CD8+ T cells producing IFN-γ, granzyme B, and perforin but also decreased (b) the amount of IFN-γ, granzyme B, and perforin production on a per cell basis, as indicated by comparative geometric mean of fluorescence (GMEAN) ± SEM of three independent experiments. Final GMEAN values are the result of the ratio between the GMEAN obtained with the experimental antibody and the isotype control.
