Effect of LPS on [ ³ H]saquinavir accumulation and inflammatory mediator release by HAPI microglia cells. (A) Following 24 hours of incubation at increasing LPS concentrations (1 to 10 ng/ml) (black bars), [³H]saquinavir (50 nM) accumulation in HAPI cells was determined at one hour (37°C) using scintillation counting. The accumulation was dose-dependent at concentrations higher than 2.5 ng/ml LPS. (B) Accumulation of [³H]saquinavir (50 nM) for one hour following addition of LPS into the transport buffer. Each data point represents mean ± SE of at least three separate experiments with three replicates included per experiment; *P < 0.05, significantly different from control.
