Lipopolysaccharide (LPS) suppressed tissue inhibitor of metalloproteinase (TIMP)-2 expression in BV2 cells and primary microglia. BV2 cells (A) or rat primary microglia (B) were treated with LPS (100 or 10 ng/ml), and total RNA was isolated at indicated times after LPS treatment. The mRNA expression of TIMP-2 was measured by RT-PCR and normalized to GAPDH expression. Representative gels are shown at the top, and quantification of three independent experiments are shown at the bottom panel. (C,D) Western blot analysis was performed using cell lysates or conditioned medium of BV2 cells treated with LPS (100 ng/ml). Levels of TIMP-2 protein expression were normalized using β-actin and were expressed as relative fold changes in comparison with control samples.
