Effect of P2x7 deficiency on splenic T-cell cytokine production. P2x7 and wildtype C57BL6 mice were immunized to develop EAE as above, and at 4 days after the booster immunization, splenic T-cells isolated and cultured in the presence (filled bars) or absence (open bars) of 20 μg/ml MOG peptide, and either 0, 50, or 100 μM BzATP. After 24 hr, levels of IFNγ and IL-17 in the culture media were determined by specific ELISA. Cytokine production is shown for WT (A, C) and P2x7 null (B, D) cells.
