cPLA 2 α is increased in the ischemic hemisphere following focal ischemia. A: A representative Western blot of cPLA2α+/+ brain protein fractions (25 μg each lane) from the non-ischemic (contralateral) and ischemic (ipsilateral) hemispheres. The same membrane was exposed to antibody and developed sequentially for phospho-cPLA2α, cPLA2α, and actin. The panel on the left represents positive controls-recombinant human cPLA2α (rh cPLA2α) and mouse spleen protein-and a negative control, cPLA2α-/- brain protein. The antibody directed against cPLA2α recognizes both the phosphorylated and nonphosphorylated cPLA2α.
