Effects of CYT and quercetin on LPS-induced IL-32 production. PBMCs (2 × 105) were treated with CYT, quercetin, or caspase-1 inhibitor (10 μM) for 2 h and then stimulated with LPS (10 ng/ml) for 24 h. Cytokine concentrations were measured in cell supernatants using the ELISA method (A). PBMCs (2 × 106) were treated with CYT (1 mg/ml) or quercetin (0.1 mM) for 2 h and then stimulated with LPS (10 ng/ml) for 8 h. Messenger RNA was measured using RT-PCR method. Results are representative of three independent experiments and band intensities correspond to levels of IL-32/GAPDH rate (B).*P < 0.05, significantly different from the unstimulated cells. **P < 0.05 compared to LPS alone.
