From: Inhibitory effect of a tyrosine-fructose Maillard reaction product, 2,4-bis(p-hydroxyphenyl)-2-butenal on amyloid-β generation and inflammatory reactions via inhibition of NF-κB and STAT3 activation in cultured astrocytes and microglial BV-2 cells

Figure 5

Effect of 2,4-bis( p -hydroxyphenyl)-2-butenal on expression of BACE1, C99 and Aβ 42 , secretase activity and Aβ 42 level. Expressions of BACE1, Aβ 42 and C99 were detected by western blotting using specific antibodies in astrocytes (A) and microglial BV-2 cells (E). Each blot is representative of three experiments. β-Actin protein was used here as an internal control. *p < 0.05 indicates significantly different from the LPS-treated group. Co-treatments with 2,4-bis(p-hydroxyphenyl)-2-butenal and LPS for 24 h were used. Media were collected to determine Aβ42 secretion by ELISA from astrocytes (B) and microglial BV-2 cells (F). Values represent the mean ± S.E. of three independent experiments with triplicate. The activities of β-secretase and of γ-secretase were assessed using commercially available assay kits as described in Methods (C and D). Values represent mean ± S.E. for three independent experiments performed in triplicate. * indicates significantly different from LPS treated group (p < 0.05).
