S1P 5 is expressed in the human cerebrovasculature. Capillaries in white matter from non-neurological human controls show strong S1P5 expression. Human S1P5 expression was determined with either the Santa Cruz (a) or the Imgenex (b) antibody both showing similar staining patterns. Colocalization studies with the EC marker CD31 (c) confirmed that ECs express significant levels of S1P5 (d) in human brain capillaries (e). S1P receptor RNA expression levels of hCMEC/D3 cells were determined by qPCR. S1P1 expression was most abundant while there was no detectable level of S1P4 expression. Receptor expression was detected in the following order: S1P1 > S1P5 > S1P3 > S1P2 where S1P1 expression level was set at 100% (f). RNA encoding for S1P4 was undetectable (n.d.). Data represent ± SEM of three independent experiments.
