S1P 5 knockdown abolishes the immune quiescent state of the brain endothelial cell barrier. (a) RNA expression of proinflammatory cytokines and chemokines were assayed by qPCR. RNA expression of the chemokines MCP-1 and IL-8 are increased by 78% (control: 100.0% ± 5.6; S1P5 knockdown 178.1% ± 9. 9, n = 3) and 171% (control: 100.0% ± 6.7; S1P5 knockdown 271.4% ± 16.3, n = 3) respectively in S1P5-deficient cells compared to mock cells. (b) Levels of the endothelial cytokines were increased upon S1P5 deficiency, IL-1β by 442% (control: 100.0% ± 7.1; S1P5 knockdown 541.5% ± 58. 5, n = 3) and TNF-α by 138% (control: 100.0% ± 22.9; S1P5 knockdown 238.4% ± 24.7, n = 3).
