Effect of MOG-DNA vaccination in T and B cell immune responses. (A) Splenocytes isolated at day 14 p.i. from mice challenged for EAE induction with MOG35-55 and previously vaccinated with DNA (MOG or pCi) were incubated in the presence (stimulated) or absence (control) of MOG35–55 (10 μg/ml) for 48 h. Cytokine production in cell culture supernatants was determined by flow cytometry. Bars indicate mean values (SEM) in pg/ml of five mice per group. *p = 0.016 for IFN-γ and p = 0.013 for IL-17 (Student’s t-test). Results shown are representative of two independent experiments. (B) Serum samples from EAE mice treated with MOG-DNA or pCi were obtained at day 30 p.i., and anti-MOG35-55 IgG titers were determined by ELISA. Error bars indicate mean values and SEM. Data represent a pool of three different experiments.
