LRRK2 kinase inhibition attenuates Tat-induced microglial phagocytosis of uncharged and carboxylated latex beads. We treated BV-2 cells with saline or Tat for 12 hours ± LRRK2i and then exposed the cultures to uncharged latex beads for 1.5 hours. (A) LRRK2i significantly decreased Tat-induced phagocytosis of uncharged latex beads (n = 3 per condition). We then treated BV-2 cells with Tat for 6 hours and exposed them to carboxylated, fluorescent latex beads for 45 minutes. Cells treated with Tat (C) contain many more latex beads (red) than saline treated cells (B). Treatment with LRRK2i in both the saline (D) and Tat (E) conditions decreased the number of engulfed beads (*P <0.05, ***P <0.001, one way ANOVA, Newman-Keuls post-test).
