Effects of MIP-2γ overexpression on astrocyte glutamate uptake. Results are expressed as the ratio of [3H]glutamate uptake of transfected cultures over the value of the control cultures ± SEM. Glutamate uptake capacity was decreased in astrocytes transfected with pAAV-MIP-2γ-hrGFP plasmids (MIP-2γ) compared with untreated cultures (control) or transfected with pAAV-IRES-hrGFP (mock) vector. Astrocytes co-transfected with pAAV-MIP-2γ-hrGFP plasmids and siRNA-1 (MIP-2γ + siRNA-1), but not with pBS/U6-con (MIP-2γ + BS/U6-con), showed partially restored glutamate uptake. n = 3, *P <0.05; **P <0.01 significantly different from the value obtained in control (ANOVA followed by a Bonferroni test).
