Role of FoxO1 in lithium regulates TLR4 signaling. (a) Suppression of FoxO1 expression by FoxO1 siRNA. The primary microglial cells were transfected with TLR4 siRNA (100 nM) or control (siControl) for 48 hours. (b) Western blotting analysis of the expression of TLR4, p-PI3K and p-Akt in lipopolysaccharide (LPS)-stimulated primary microglia after transfection with FoxO1 siRNA (100 nM) and/or LiCl (3 mM). The levels of p-PI3K, p-Akt and TLR4 (c) were quantified and normalized with their respective total PI3K, Akt or GAPDH levels. Each value was then expressed relative to the one treated with medium, which was set as 1.
