Locally produced IL-10 might be responsible for anti-inflammatory response . (A) IL-10 gene expression at 3 days post inoculation (dpi) and 7 dpi in Treg-depleted (diphtheria toxin; DT) and non-depleted (PBS) mice was assessed by RT-PCR. The bar graphs represent mean ± SD of fold change in gene expression obtained by analyzing four mice from each group. (B) Double-immunofluorescence stainings of coronal brain sections using antibodies against IL-10 (red) in combination with Iba-1 (microglia), glial fibrillary acidic protein (GFAP) (astrocytes) or CD3+ (T cells) at 7 dpi in Treg-depleted mice show that microglia are the major source of IL-10.
