Celastrol suppressed the expression of proinflammatory genes in LPS-stimulated ARPE19 and RAW264.7 cells. ARPE19 (a) or RAW264.7 cells (b) were pretreated with celastrol at indicated concentrations for 30 min, which was followed by LPS incubation at concentrations of 1 μg/ml and 5 ng/ml, respectively. Cells were harvested 6 h later for RNA extraction. Real-time PCR analyses were subsequently performed to examine the expression of IL1β, Ccl2, TNFα, and ICAM-1. Data were expressed as mean ± S.E.M.
