Cerebral oligodendroglial differentiation is not affected by direct FXR activation or supernatant of FXR-activated BMMs. Cerebral oligodendrocytes show no differences in Mbp expression when incubated for 24 or 48 h with 1 μM or 10 μM GW4064 compared to control cells treated with solvent (a). Numbers of PDGFRα+ OPCs and MBP+ oligodendrocytes are not altered due to GW4064 treatment after 48 h (b, c ). Supernatants of BMMs reduce Mbp gene expression (d) and the percentage of MBP+ oligodendrocytes after 48 h of differentiation, but this effect is independent of GW4064 (e, f ).
