Treatment with CSF1R inhibitor PLX5622 alters brain gene expression. Mice received chow containing PLX5622 for 3 weeks and mRNA was isolated from whole brain. a RT-PCR showed marked reduction in several microglial transcripts, indicating substantial microglial depletion. Other neuroimmune transcripts were also assessed. b The effects of PLX5622 on non-microglial transcripts were also assessed. Note that PLX5622 had little to no effect on neuronal, oligodendrocyte, or endothelial transcripts, but increased the astrocyte marker GFAP.
