Cyclooxygenase enzymatic activity in primary rat microglia cells is decreased by AM 404. For COX-1 activity assay (a), cells were treated with different concentrations of AM 404 (0.1, 1, 5, or 10 μM) for 15 min before the addition of 15 μM of arachidonic acid. PGE₂ in the supernatants was measured after additional 15 min. For COX-2 (b), cells were stimulated for 24 h with LPS (10 ng/mL) and then treated with different concentrations of AM 404 for 15 min. After this incubation time, 15 μM of arachidonic acid was added and PGE₂ in the supernatants was measured as described in “Methods.” Data is expressed as mean ± SEM of at least three new cultures/group. *p < 0.05, **p < 0.01, and ***p < 0.001 with respect to control (one-way ANOVA followed by the Newman-Keuls post-test)
