Tenuigenin inhibits NLRP3 inflammasome activation induced by ATP in BV2 microglia. ae Immunoblot analysis of IL-1β in culture supernatants of LPS-primed BV2 treated for 3 h with various doses of tenuigenin and then stimulated with ATP (2.5 mM) for 30 min, and immunoblot analysis of NLRP3, the precursors of IL-1β (pro-IL-1β) and caspase-1 (pro-caspase-1), cleaved caspase-1 (p20), and IL-1β in lysates of those cells. f ELISA of matured IL-1β in supernatants from LPS-primed BV2 treated for 3 h with various doses of tenuigenin and then stimulated with ATP. g Cells were stained with DCFDA labeling and the quantitative analysis of ROS levels.
