CCL2/CCR2 signalling in monocytes/macrophages plays a crucial role in VCR pain in CX3CR1-deficient mice. (1) Downregulation or silencing of CX3CR1 in monocytes results in upregulation of CCR2 via p38 MAP kinase signalling. (2) Monocytes infiltrate through the endothelium into peripheral nerve tissue in a CCL2/CCR2-dependent manner. (3) Cytokines such as TNFα and IL1β are released downstream of CCR2 signalling in monocytes. (4) TRPV1 channels mediate noxious signalling following TNFα receptor activation in sensory neurons
