Knock-out of CIITA attenuates α-syn-induced MHCII expression and prevents neurodegeneration in vivo. a Representative images of AAV2-SYN-induced MHCII expression (DAB, brown) in the ventral midbrain of WT and CIITA −/− mice. Both groups received unilateral AAV2-SYN injection into the ipsilateral SNpc. b Quantification of MHCII expression. Sections were analyzed by tiling the entire section and calculating the midbrain mean gray value of both contralateral and ipsilateral hemispheres in ImageJ, adjusting for background. Ipsilateral and contralateral values were used to determine fold induction of MHCII in the ipsilateral side. Data represent the mean MHCII induction, with each data point representing an individual animal. Three brain sections were analyzed per animal, with six animals per group.
