ALX treatment inhibits Th1 and Th17 responses in peripheral lymphoid organs of EAE mice. On day 25 post-induction, 5–6 mice in each group were sacrificed. The total numbers of splenic mononuclear cells and CD4+ T cells were calculated. Their splenic Th1 and Th17 cells were characterized by flow cytometry. The levels of splenic IFN-γ and IL-17 were determined by ELISA, and IFN-γ, IL-17, T-bet, and RORγt were determined by quantitative RT-PCR. Data are representative images or expressed as the means ± SEM of each group from three separate experiments. a Total number of splenic mononuclear cells and CD4+ T cells. b Flow cytometry analysis of the percentages of splenic Th1 and Th17 cells in the vehicle- and ALX-treated mice. c ELISA analysis of IFN-γ and IL-17 production in the supernatants of cultured splenocytes. d The relative levels of IFN-γ, T-bet, IL-17, and RORγt mRNA transcripts in the spleen of individual mice. ※p < 0.05, *p < 0.01, #p < 0.001
