A schematic illustration of the proposed mechanism for H2R/H3R to alleviate microglia-induced neuroinflammation. LPS or stress activates TLR4 and induces microglia to release inflammatory cytokines. PI3K/AKT/FoxO1 axis, as the downstream of TLR4, provides a self-limiting mechanism by which microglia avoid inappropriate long-term overactivation. H2R/H3R can activate PI3K/AKT/FoxO1 signaling, trigger the phosphorylation and nuclear exclusion of FoxO1, and inhibit microglia TLR4 expression and TLR4-mediated inflammatory cytokine release
