Cerulenin blocks LPS-induced secretion of neurotoxins by BV2 cells. a BV2 cells were pretreated for 1 h with vehicle (0.01% DMSO), 10 μM cerulenin, or 5 μM BAY11-7082, followed by treatment for 24 h with 0 (CTL:1x PBS) or 100 ng/ml LPS to obtain conditioned media. HT22 cells grown in regular medium were switched to the conditioned media. After 24 h, cell viability was measured using MTT assay. b HT22 cells were cultured in various microglia-conditioned media (MCM) shown with or without addition of 10 μM cerulenin as indicated. Cell viability was measured as above.
