STAT6 is required for PPARĪ³ activation. BV-2 cells were treated with leflunomide (10 Ī¼M) or GW9662 (1 Ī¼M) 30 minutes prior to administration of RvD1 (100 nM), followed by incubation with IL-4 (10 ng/ml) for 120 minutes. PPARĪ³ protein level in nucleus (A) and DNA binding activity (B) were analyzed using Western blot or electrophoretic mobility shift assay (EMSA). A representative result from three independent experiments is shown. Quantification of PPARĪ³ was normalized by lamin B1. Data are presented as meanā€‰Ā±ā€‰SEM for three independent experiments. Asterisks indicate statistically significant difference (*Pā€‰<ā€‰0.05, **Pā€‰<ā€‰0.01).
