From: Cerebellar Purkinje cells incorporate immunoglobulins and immunotoxins in vitro: implications for human neurological disease and immunotherapeutics

Immunoglobulin

Immunoglobulin localization within Purkinje cellsa

Immunoglobulin clearance from Purkinje cellsb

IgGsc

4 hours: Purkinje cell processes. Rare Purkinje cell bodies

6-8 hours: Purkinje cell processes and cytoplasm

24-48 hours: Most Purkinje cells (processes, cytoplasm, and nuclei)

24-48 hours

Normal Rat IgM

4 hours: Purkinje cell processes. Rare Purkinje cell bodies

6-8 hours: Purkinje cell processes and cytoplasm

24 hours: Most Purkinje cells (processes, cytoplasm, and some nuclei)

48 hours: Most Purkinje cells (processes, cytoplasm, and nuclei)

24-48 hours

  1. a. Purkinje cell death was observed in <5% of cells through 120 hours of incubation as indicated by intracellular presence of the cell death marker, SYTOX green. Cell death was equivalent to that seen in cultures incubated without immunoglobulin.
  2. b. Following transfer of cultures to media not containing the immunoglobulin
  3. c. IgGs: normal rat IgG, GK1.5 monoclonal rat anti-CD4+ antibody, normal human IgG, Cy5-conjugated donkey anti-human IgG, FITC-conjugated donkey anti-human IgG. Cy5-conjugated donkey anti-human IgG and FITC-conjugated donkey anti-human IgG were studied both in real time in living cultures and following fixation.
