Spinal patterning of peptidergic (CGRP positive) and non-peptidergic (Isolectin B4 binding) terminals in the lumbar spinal cord of control immunized and SJL-PLP 139-151 immunized mice over the time course of the EAE. (A) Examples of CGRP expression at 2 to 3 days following immunization (pre-timepoint), in the onset phase, in the peak phase and in the chronic phase of the disease (day 35 to 40 after immunization). (B) Examples of Isolectin B4 (IB4) binding at 2 to 3 days following immunization (pre-timepoint), in the onset phase, in the peak phase and in the chronic phase of the disease (day 35 to 40 after immunization). There was an increase in IB4 positive binding throughout the spinal cord in SJL-EAE mice in the onset and peak phase of the disease. (C) Co-labeling of anti-GFAP with IB4 revealed no overlay. (D) Co-labeling of anti-Iba1 with IB4 showed strong colocalization. Examples are indicated with arrows. Scale bars represent 1 mm in panel A and B and 50 μm in panel C and D. CGRP, calcitonin gene-regulated peptide; EAE, experimental autoimmune encephalomyelitis; IB4, nonpeptidergic isolectin B4.
