T cell responses in the spleens following WNV NY99 and WNV Eg101 infection. Spleens were harvested after extensive perfusion of mock, WNV NY99, and WNV Eg101 infected mice at day 8 after inoculation. Splenocytes were isolated and stimulated ex vivo for 6 h with an immunodominant NS4B peptide as described in the materials and methods. Cells were co-stained for CD8 and intracellular IFN-γ and TNF-α and analyzed by flow cytometry. a Total number of CD8+ TNF-α+ cells, CD8+ IFNγ+ cells, and CD8+ TNF-α+ IFNγ+ cells.
