The NF-κB inhibitor Ro-106-9920 blocks LPS-induced up-regulation of TLR2 gene expression in rat cortical astrocytes. Purified (L-LME-treated) astrocytes were first incubated 30 min with 10 μM Ro-106-9920 (‘Ro-106’) in serum-containing medium, followed by addition of LPS to a final concentration of 100 ng/ml LPS. After a further 6 h, cells were collected and processed for TLR2 mRNA expression by RT-PCR.
