SIRT1 is a negative regulator of Foxp3 expression in human T cells. SIRT1 knockdown in primary human CD4+ T cells of healthy volunteers was performed by specific siRNA, and Foxp3 mRNA expression and Treg differentiation was analyzed after incubation under Treg skewing conditions for 4 days. a Successful transfection of siRNA was confirmed by qPCR (left panel) and Western Blot analysis (right panel).
