Effect of caveolin-1 (cav-1) on the blood-brain barrier (BBB). Cav-1 increases the BBB permeability via caveolae-based transcytosis and translocation of tight junction (TJ) protein. Cav-1 inhibits matrix metalloproteinase-9 (MMP-9) which disrupts TJ proteins and basement membrane under ischemic stroke condition, while cav-1 appears to promote MMP-9 upregulation by tPA. However, cav-1 (−/−) mice demonstrated higher MMP activity and BBB permeability than cav-1(+/+) mice in a focal cerebral ischemia-reperfusion model, which means that cav-1 may mainly protects BBB integrity after IS.
